We see you've tried removing your case, restarting the phone, testing in Safe Mode, and running diagnostics.
The last step in troubleshooting would be a factory reset. Be sure to do a complete backup first. https://www.samsung.com/us/support/answer/ANS10002030/
If the area is still not responsive with the S Pen, we recommend having the phone evaluated by an authorized service center. Fold & Flip Phones@Lorek Do you have a case on your Fold 6? Does it possibly have a magnet in the case?
A magnetic case can affect S Pen operation.
Remove the case and check for proper operation.

Fold & Flip PhonesYou may have a 3rd party software conflict. You can try booting in safe mode. If it works on while booted in safe mode, it is definitely a conflict with some 3rd party software.
